The compound was developed in the late 1980s and early 1990s by academic researchers investigating melanocortin signaling and pigmentation. Early work explored whether synthetic analogs could reproduce effects of the natural hormone under controlled conditions. The molecule never advanced through the full regulatory pathway required for approval as a medicine. From the mid-2000s onward it appeared in unregulated consumer markets, often distributed through informal channels. That gap between research origins and commercial availability shapes how the compound is discussed today.
Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.
Published research on the compound remains limited. Much of the human data comes from small, early-stage studies rather than large controlled trials, and several questions about effects and variability between individuals remain open. Investigators have examined receptor activity, pigment pathways, and related physiological responses in laboratory and animal models. Findings from those models do not automatically translate to human outcomes. Reviews frequently note the scarcity of rigorous clinical evidence and call for better-characterized study material.
Because the substance circulates mainly through informal markets, verification is a recurring theme in technical discussion. Independent analyses have found that labeled content and actual content can diverge, and that purity varies between samples. Analytical laboratories use reversed-phase chromatography to separate components and mass spectrometry to confirm identity. Isotope-labeled internal standards improve quantification in complex matrices. Such methods describe what a sample contains but say nothing about its sterility, lawful status, or suitability for any use. Open questions remain about how consistently testing is applied across the supply chain.
Regulatory treatment of this peptide varies by country. It holds no marketing authorization as a medicine in the United States, the European Union, or most other jurisdictions. Some countries classify products containing it as prescription-only or unlicensed medicines, which restricts lawful supply. Authorities have issued public notices warning that unregulated products may contain undeclared or incorrect ingredients. The molecule also appears on prohibited lists for competitive sport. These measures address supply oversight rather than any approved therapeutic role.

No regulatory authority has approved melanotan-2 for human use, and several countries classify it as a prescription-only or controlled substance, which restricts lawful supply. Material sold online is generally labelled as a research chemical and is not required to meet pharmaceutical standards of identity or purity. Published human data consist mainly of small uncontrolled studies, case reports and adverse-event notifications, so the evidence base is descriptive rather than confirmatory. Whether repeated melanocyte stimulation alters long-term naevus behaviour remains an open question that no completed trial has resolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous tridecapeptide that regulates pigment production. Two modifications distinguish it from the natural hormone: norleucine replaces methionine at the N-terminus, which limits oxidation, and a D-phenylalanine substitution raises receptor affinity. The ring is closed through an aspartate-lysine lactam bridge, giving the molecule a constrained conformation. The free base has a molecular mass near 1024 daltons, and commercial material is usually supplied as an acetate salt. It appears in the literature as a research peptide rather than an approved therapeutic agent.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Wolverine's mental health struggles have often been analyzed by academics and psychologists, cited as part of the character's appeal with audiences. In addition to his comic appearances, Wolverine has been depicted in a wide range of adapted and spin-off media, including television, video games, and film. Wolverine's first major appearance outside of comics was in X-Men: The Animated Series (1992–97), voiced by Cal Dodd, which helped popularize the character among the general public; Dodd reprises his role in the revival series X-Men '97 (2024–present). Actor Hugh Jackman has portrayed the character in ten films, beginning with X-Men (2000) and most recently in Deadpool & Wolverine (2024).
A very important modification of eukaryotic pre-mRNA is RNA splicing. The majority of eukaryotic pre-mRNAs consist of alternating segments called exons and introns. During the process of splicing, an RNA-protein catalytical complex known as spliceosome catalyzes two transesterification reactions, which remove an intron and release it in form of lariat structure, and then splice neighbouring exons together. In certain cases, some introns or exons can be either removed or retained in mature mRNA. This so-called alternative splicing creates series of different transcripts originating from a single gene. Because these transcripts can be potentially translated into different proteins, splicing extends the complexity of eukaryotic gene expression and the size of a species proteome. Extensive RNA processing may be an evolutionary advantage made possible by the nucleus of eukaryotes. In prokaryotes, transcription and translation happen together, whilst in eukaryotes, the nuclear membrane separates the two processes, giving time for RNA processing to occur.

== Clinical significance == NNMT is highly expressed in the human liver. N-methylation is one method by which drug and other xenobiotic compounds are metabolized by the liver. NNMT expression in adipose tissue is associated with obesity and insulin resistance. Contrary to the negative effects of increased NNMT in adipose tissue, increased NNMT in liver is associated with a better metabolic profile, namely reduced serum triglycerides and free fatty acids. In adipose tissue, NNMT can lead to methylation depletion, whereas because of the many methylation enzymes in the liver NNMT has a negligible effect on liver methylation. But in the liver, the 1-methylnicotinamide produced by NNMT degradation of nicotinamide increases sirtuin 1 (SIRT1) by inhibiting degradation of that protein. Overexpression of SIRT1 in mice has been shown to reduce insulin and fasting glucose, as well as increased metabolism and physical function. Abundant availability of nicotinamide leads to depletion of both nicotinamide adenine dinucleotide (NAD+) and SAM-e, resulting in liver steatosis and fibrosis, causing the progression from non-alcoholic fatty liver disease (NAFLD) to non-alcoholic steatohepatitis (NASH). Human embryonic stem cells expression of NNMT is believed to help maintain the cells in a naive state. NNMT expression is significantly upregulated in many cancers, including pancreatic cancer where levels of NNMT enzyme correlate with increased risk of death. The cause of these correlations has not been established, but may be related to the fact that NNMT enzyme is an inhibitor of DNA repair.

Enishi (Hokkaido) Teishu (Honshu) Iyo (Shikoku) Tsukushi (Kyushu) Ryugu (Okinawa) Like every other nation in the world, Yamato has its own Agents of the Four Seasons, each of whom has the duty to manifest their season in every part of the land. The Agents of Spring and Summer begin their journey on the island of Ryugu then move north, while the Agents of Autumn and Winter begin their journey on the island of Enishi, then head south. The role of "Agent" is bestowed by the deities of the seasons based on the physical, mental and spiritual eligibility of a blood descendant of the first Agent, and the chosen individual cannot in any way oppose their destiny. The bestowal occurs when the previous Agent dies or ceases to be eligible for the role. It is possible for an Agent, due to insufficient preparation or drastic events, to be unable to manifest their season, consequently extending the duration of the previous one. This is the case of the Agent of Spring, Hinagiku Kayo, a 16-year-old girl who was kidnapped by the Insurgents at the age of just 6. As a result, spring hasn't manifested in Yamato for 10 years, and winter, instead of lasting 3 months like the other seasons, has lasted 6 months during this period of time. However, the young Hinagiku has made her return and is now preparing to bring her season back to the nation, accompanied by her guard, Sakura Himedaka. It all begins on February 10th of the year 20 of the Reimei era, on the island of Ryugu.
A separate co-op campaign is also available for up to four players, following a parallel plot where the players control a group of police officers that also get trapped in the nightmare world while investigating Simon. Additionally, there is also a co-op survival mode, where players fight several waves of enemies for a set period of time. Some days before Cry of Fear's anniversary, Valve released a Half-Life update for Linux compatibility, making changes in the folders and engine. This update made several Half-Life mods, including Cry of Fear, incompatible with the base game. Team Psykskallar decided that, since no more could be done for the mod itself, they would finish a standalone version. Confusion due to Valve regarding Cry of Fear's status as freeware caused the game to be delayed until April 25, 2013.
Some organizations—but fewer than in the past—require that patients spend a certain period of time living in their desired gender role before starting hormone therapy. This period is sometimes called real-life experience (RLE). In Sweden, for instance, patients seeking to access gender affirming healthcare must first undergo extended evaluations with psychiatric professionals, during which they must—without any form of medical transition—successfully live for one full year as their desired gender in all professional, social, and personal matters. Gender clinics are recommended to provide patients with wigs and breast prostheses for the endeavor. The evaluation additionally involves, if possible, meetings with family members and/or other individuals close to the patient. Patients may be denied care for any number of "psychosocial dimensions", including their choice of job or their marital status. Transgender and gender non-conforming activists, such as Kate Bornstein, have asserted that RLE is psychologically harmful and is a form of "gatekeeping", effectively barring individuals from transitioning for as long as possible, if not permanently. In September 2022, the World Professional Association for Transgender Health (WPATH) Standards of Care for the Health of Transgender and Gender Diverse People (SOC) Version 8 were released and removed the requirement of RLE for all gender-affirming treatments, including gender-affirming surgery.
In 1027 and 1044, Patriarch Poppo of Aquileia, who rebuilt the cathedral of Aquileia, entered and sacked neighbouring Grado, and, though the Pope reconfirmed the Patriarch of the latter in his dignities, the town never fully recovered, though it continued to be the seat of the Patriarchate until its formal transference to Venice in 1450. In the fourteenth century, the Patriarchal State reached its largest extension, stretching from the Piave river to the Julian Alps and northern Istria. The seat of the Patriarchate of Aquileia had been transferred to Udine in 1238, but returned to Aquileia in 1420 when Venice annexed the territory of Udine. In 1445, the defeated patriarch Ludovico Trevisan acquiesced in the loss of his ancient temporal estate in return for an annual salary of 5,000 ducats allowed him from the Venetian treasury. Henceforth only Venetians were allowed to hold the title of Patriarch of Aquileia. The Patriarchal State was incorporated into the Republic of Venice with the name of Patria del Friuli, ruled by a provveditore generale or a luogotenente living in Udine.
=== Ethidium bromide === Ethidium bromide intercalates and stains DNA, providing a fluorescent red-orange stain. Although it will not stain healthy cells, it can be used to identify cells that are in the final stages of apoptosis – such cells have much more permeable membranes. Consequently, ethidium bromide is often used as a marker for apoptosis in cells populations and to locate bands of DNA in gel electrophoresis. The stain may also be used in conjunction with acridine orange (AO) in viable cell counting. This EB/AO combined stain causes live cells to fluoresce green whilst apoptotic cells retain the distinctive red-orange fluorescence.

== Pronunciation == Pronunciation follows convention outside the medical field, in which acronyms are generally pronounced as if they were a word (JAMA, SIDS), initialisms are generally pronounced as individual letters (DNA, SSRI), and abbreviations generally use the expansion (soln. = "solution", sup. = "superior"). Abbreviations of weights and measures are pronounced using the expansion of the unit (mg = "milligram") and chemical symbols using the chemical expansion (NaCl = "sodium chloride"). Some initialisms deriving from Latin may be pronounced either as letters (qid = "cue eye dee") or using the English expansion (qid = "four times a day").
In Bangladesh, a member of parliament is an individual who serves in the unicameral Jatiya Sangsad or House of the Nation. Members of the Jatiya Sangsad are elected at a general election, usually held once every five years unless Parliament is dissolved sooner by the president on the advice of the prime minister. Under the Constitution of Bangladesh, an individual is required to be a citizen of Bangladesh and must have attained the age of 25 years in order to qualify for election to Parliament. The Parliament consists of 300 directly elected members from general seats elected by use of first past the post who represent single-constituencies, while 50 seats are reserved exclusively for women and are allocated on a proportional basis. After an election, the Election Commission allocates reserved seats to parties based on the number of general seats they won. A party then presents a list of candidates, each requiring a presenter and a seconder. If the number of candidates presented and seats allocated is equal, then there is no election and the reserved seats are filled in accordance with the candidate lists prepared by parties. In the event there are more candidates than seat allocations, the 300 MPs elected from general seats vote through use of the single transferable vote system to determine the reserved seats. In reality, there has never been an election for reserved seats as parties have never nominated more candidates than they have been allocated. In order to form a Government, a political party or alliance usually requires a simple majority in Parliament.

Cave 17 (Cave of the Bodhisattva Vault) is large "central pillar" cave located near Cave 14, and high up on the rock to the right of Cave 8. It was probably the main cave of the group of caves from 14 to 19, which also includes several undecorated living quarters or viharas, and was dedicated to religious services. Su Bai dates Cave 17 to the Second Stage (395–465 CE -+65), together with Cave 14. Luo and Wang date it to 465 CE +-65. Rhie attributes Cave 17 to circa 500, based on stylistic considerations. It is a "central pillar" cave, with a small, very colorful, square cella (3.80x3.90 meters; 12 x 13 feet) with a vaulted ceiling, a central pillar with two side corridors, and a back room. These caves were possibly small chapels to nearby viharas. In Cave 17, on the lower left panel of the entrance wall, appeared a Royal family, composed of the King, Queen and two young Princes. They are accompanied by monks, and men in caftan. The relief is now in the Hermitage Museum. The King wears a crown and a triple halo, with Sasanian-type royal ribbons. He wears a long white caftan decorated with small diamond designs, and has long boots. His right hand is in front of his chest, holding an incense lamp, and he holds an akinakes sword and a red bag in the left hand. The end of a long knight's sword is visible behind the first boot. The king can be identified as a Tocharian king of Kucha. His Queen wears a long robe, and his two sons, Princes, wear ornate caftan and are fair-haired.
Calcined uranium yellowcake, as produced in many large mills, contains a distribution of uranium oxidation species in various forms ranging from most oxidized to least oxidized. Particles with short residence times in a calciner will generally be less oxidized than those with long retention times or particles recovered in the stack scrubber. Uranium content is usually referenced to U3O8, which dates to the days of the Manhattan Project when U3O8 was used as an analytical chemistry reporting standard. Phase relationships in the uranium-oxygen system are complex. The most important oxidation states of uranium are uranium(IV) and uranium(VI), and their two corresponding oxides are, respectively, uranium dioxide (UO2) and uranium trioxide (UO3). Other uranium oxides such as uranium monoxide (UO), diuranium pentoxide (U2O5), and uranium peroxide (UO4·2H2O) also exist. The most common forms of uranium oxide are triuranium octoxide (U3O8) and UO2. Both oxide forms are solids that have low solubility in water and are relatively stable over a wide range of environmental conditions. Triuranium octoxide is (depending on conditions) the most stable compound of uranium and is the form most commonly found in nature. Uranium dioxide is the form in which uranium is most commonly used as a nuclear reactor fuel. At ambient temperatures, UO2 will gradually convert to U3O8. Because of their stability, uranium oxides are generally considered the preferred chemical form for storage or disposal.

It is a synthetic seven-amino-acid peptide modeled on alpha-melanocyte-stimulating hormone. It carries two non-natural substitutions and a cyclic bridge that increase its stability relative to the natural hormone. It circulates as a research chemical and is not an approved medicine.
Melanotan-1, also called afamelanotide, is a shorter linear analog with a different amino acid sequence and no lactam ring. It has been evaluated in formal clinical programs, while melanotan-2 has not. The two are distinct molecules and are not interchangeable.
No. Bremelanotide is a related but distinct cyclic peptide that reached approved status for one specific clinical indication. Melanotan-2 is a separate molecule with its own sequence and properties. Shared ancestry in the melanocortin family does not make them the same substance.
Only a small number of early-stage human studies have been reported, and most were limited in size and duration. No large late-stage program has established a general efficacy or safety profile. The evidence base is therefore thin compared with approved medicines.
