Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.
The compound emerged from research programs in the 1980s that examined analogues of alpha-melanocyte-stimulating hormone for pigmentation and photoprotection. Investigators modified the native sequence to extend activity duration and potency. A related analogue, afamelanotide, was developed within the same broad line of inquiry and eventually gained approval in certain jurisdictions for a rare light-sensitivity condition. Melanotan-2 itself did not progress through the same regulatory route and has no approved therapeutic indication.
Receptor studies place melanotan-2 among non-selective melanocortin agonists, binding MC1R, MC3R, MC4R and MC5R rather than a single subtype. Activation of MC1R on cutaneous melanocytes raises tyrosinase activity and shifts pigment synthesis toward eumelanin, which is darker and more photostable than pheomelanin. Central receptors, particularly MC4R, are associated with appetite suppression and with reported effects on sexual function. Because subtype selectivity is low, the same molecule engages pigment, metabolic and vascular pathways at once, and this breadth is a common explanation offered for the range of adverse events described in user reports.
No regulatory authority has approved melanotan-2 for human use, and several countries classify it as a prescription-only or controlled substance, which restricts lawful supply. Material sold online is generally labelled as a research chemical and is not required to meet pharmaceutical standards of identity or purity. Published human data consist mainly of small uncontrolled studies, case reports and adverse-event notifications, so the evidence base is descriptive rather than confirmatory. Whether repeated melanocyte stimulation alters long-term naevus behaviour remains an open question that no completed trial has resolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous tridecapeptide that regulates pigment production. Two modifications distinguish it from the natural hormone: norleucine replaces methionine at the N-terminus, which limits oxidation, and a D-phenylalanine substitution raises receptor affinity. The ring is closed through an aspartate-lysine lactam bridge, giving the molecule a constrained conformation. The free base has a molecular mass near 1024 daltons, and commercial material is usually supplied as an acetate salt. It appears in the literature as a research peptide rather than an approved therapeutic agent.

Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
A mammary gland is a specific type of apocrine gland specialized for manufacture of colostrum (first milk) when giving birth. Mammary glands can be identified as apocrine because they exhibit striking "decapitation" secretion. Many sources assert that mammary glands are modified sweat glands.
In this pathway, adenylate kinase combines two ADP molecules to make ATP and AMP; AMP is then deaminated, producing inosine monophosphate (IMP) and ammonia (NH3) as part of purine nucleotide cycle. Physicians may also check resting levels of creatine kinase, which are moderately increased in 90% of patients. In some, the level is increased by multitudes - a person without GSD-V will have a CK between 60 and 400IU/L, while a person with the syndrome may have a level of 5,000 IU/L at rest, and may increase to 35,000 IU/L or more with muscle exertion. This can help distinguish McArdle's syndrome from carnitine palmitoyltransferase II deficiency (CPT-II), a lipid-based metabolic disorder that prevents fatty acids from being transported into mitochondria for use as an energy source. Also, serum electrolytes and endocrine studies (such as thyroid function, parathyroid function, and growth hormone levels) will also be completed. Urine studies are required only if rhabdomyolysis is suspected. Urine volume, urine sediment, and myoglobin levels would be ascertained. If rhabdomyolysis is suspected, serum myoglobin, creatine kinase, lactate dehydrogenase, electrolytes, and renal function will be checked. Physicians may also conduct an exercise stress test to test for an inappropriately rapid heart rate (sinus tachycardia) in response to exercise. Due to the rare nature of the disease, the inappropriate rapid heart rate in response to exercise may be misdiagnosed as inappropriate sinus tachycardia (which is a diagnosis of exclusion).
After injection, inhalation, or ingestion, the first symptoms to appear are generalized muscle spasms. They appear very quickly after inhalation or injection – within as few as five minutes – and take somewhat longer to manifest after ingestion, typically approximately 15 minutes. With a very high dose, the onset of respiratory failure and brain death can occur in 15 to 30 minutes. If a lower dose is ingested, other symptoms begin to develop, including seizures, cramping, stiffness, hypervigilance, and agitation. Seizures caused by strychnine poisoning can start as early as 15 minutes after exposure and last 12–24 hours. They are often triggered by sights, sounds, or touch and can cause other adverse symptoms, including hyperthermia, rhabdomyolysis, myoglobinuric kidney failure, metabolic acidosis, and respiratory acidosis. During seizures, mydriasis (abnormal dilation), exophthalmos (protrusion of the eyes), and nystagmus (involuntary eye movements) may occur. As strychnine poisoning progresses, tachycardia (rapid heart beat), hypertension (high blood pressure), tachypnea (rapid breathing), cyanosis (blue discoloration), diaphoresis (sweating), water-electrolyte imbalance, leukocytosis (high number of white blood cells), trismus (lockjaw), risus sardonicus (spasm of the facial muscles), and opisthotonus (dramatic spasm of the back muscles, causing arching of the back and neck) can occur. In rare cases, the affected person may experience nausea or vomiting.
== Other animals == In the long-footed mammals, both the hoofed species (unguligrade) and the clawed forms which walk on the toes (digitigrade), the heel is well above the ground at the apex of the angular joint known as the hock. In plantigrade species, it rests on the ground. In birds, the heel is the backward-pointing joint which is often mistaken as the "knee" (the actual knee of birds is hidden under the plumage).

The concept of in situ in contemporary art developed in the late 1960s and 1970s, referring to works created for a specific site. Such works incorporate the site's physical, historical, political, and sociological characteristics as integral elements of their composition. This approach contrasts with autonomous artistic production, where artworks are conceived independently of their display location. Theoretical discussions, particularly in the writings and practice of French conceptual artist and sculptor Daniel Buren, have addressed the dynamic relationship between artistic intervention and its surrounding environment. The site-specific installations of Christo and Jeanne-Claude exemplify the application of in situ principles in art. Their large-scale interventions such as The Pont Neuf Wrapped (1985) and Wrapped Reichstag (1995) involved the systematic wrapping of buildings and landscape elements in fabric, temporarily transforming familiar spaces and altering public perception. Another example is the land art movement, wherein artists such as Robert Smithson and Michael Heizer integrated their works directly into natural landscapes. In contemporary aesthetic discourse, in situ describes practices that reinforce the fundamental unity between an artwork and its site.
== Applications == Desalting is used to remove salts from protein solutions, phenol or unincorporated nucleotides from nucleic acids or excess crosslinking or labeling reagents from conjugated proteins. Buffer exchange is used to transfer a protein solution into a buffer system appropriate for downstream applications such as ion exchange, electrophoresis or affinity chromatography.
Albany is in the central part of Oregon's most populated region, the Willamette Valley. The city rests along the confluence of the Calapooia and Willamette rivers, and although most of Albany falls within Linn County, a smaller portion of the city rests to the north of its downtown on the west bank of the Willamette River in Benton County. According to the United States Census Bureau, the city has a total area of 17.75 square miles (45.97 km2), of which 17.54 square miles (45.43 km2) is land and 0.21 square miles (0.54 km2) is water. Albany has 21.7 square miles (56 km2) within its urban growth boundary. Throughout the city limits and urban growth area, there are limited hills; the city is one of the lowest points along the Willamette Valley, with elevations ranging 180 to 430 feet (55 to 131 m) above sea level. The North Albany district has the most variable elevation, while the downtown and southern end of town have little elevation change throughout.
Dershowitz's campaign to acquit von Bülow was assisted by Jim Cramer and future New York attorney general and governor Eliot Spitzer, who were then Harvard Law School students. Dershowitz and his team focused on the discovery of the bag containing the syringes and insulin. Sunny's family had hired a private investigator to look into her coma. The private investigator, Edwin Lambert (an associate of the Bülows' lawyer Richard Kuh), was told by several family members and a maid that Claus had recently been seen locking a closet in the Newport home that previously was always kept open. The family hired a locksmith to drive to the mansion, with the intention of picking the closet lock to find what the closet contained. They had lied to the locksmith and told him that one of them owned the house. When the three arrived, the locksmith insisted that they try again to find the key, and after some searching, Kuh found a key in von Bülow's desk that unlocked the closet. At this point, according to the three men in the original interviews, the locksmith was paid for the trip and left before the closet was actually opened, although the men would later recant that version and insist that the locksmith was present when they entered the closet. It was in the closet that the main evidence against von Bülow was found. In 1984, the two convictions from the first trial were reversed by the Rhode Island Supreme Court. In 1985, after a second trial, von Bülow was found not guilty on all charges.
=== AFL policy === The AFL is a signatory to ASADA and WADA, and its policy on performance-enhancing drugs is based upon those organisations. For non-performance-enhancing drugs, the AFL currently has a "three-strikes" policy, under which only the player and the club doctor are aware of any positive tests until a third such test is received. After the third strike, the club is made aware of the situation and the player may face disciplinary action. This policy has faced criticism for leniency from anti-doping bodies, particularly the World Anti-Doping Agency (WADA), and as such has attracted much media scrutiny and public debate.

=== Label-free drug screening === Thermofluor has been extensively used in drug screening campaigns. Because Thermofluor detects high affinity binding sites for small molecules on proteins, it can find hits that bind to active site subsites, cofactor sites, or allosteric binding sites with equal efficacy. The method typically requires the use of screening compound concentrations at >10x the desired binding threshold. Setting 5 μM as a reasonable hit threshold consequently requires a test ligand concentration of 50 to 100 μM in the sample well. For most drug compound libraries, where many compounds are not soluble beyond ~100 μM, screening multiple compounds is consequently not feasible owing to solubility issues. Thermofluor screens do not require the development of custom screening reagents (e.g. cleavable substrate analogs), do not require any radioactive reagents, and are generally less sensitive to the effects of compounds that are chemically reactive with protein active site residues, and that consequently show up as undesirable hits in enzyme activity screens.
=== Alkanes === For linear alkanes, molecular ion peaks are often observed. However, for long chain compounds, the intensity of the molecular ion peaks are often weak. Linear fragments often differ by 14 Da (CH2 = 14). For example, hexane fragmentation patterns. The m/z=57 butyl cation is the base peak, and other most abundant peaks in the spectrum are alkyl carbocations at m/z=15, 29, 43 Da.
Target-mediated drug disposition (TMDD) is the process in which a drug binds with high affinity to its pharmacological target (for example, a receptor) to such an extent that affects its pharmacokinetic characteristics. Various drug classes can exhibit TMDD, most often these are large compounds (biologics such as antibodies, cytokines or growth factors) but also smaller compounds can exhibit TMDD (such as warfarin and CHK-336). A typical TMDD pattern of antibodies displays non-linear clearance and can be seen at concentration ranges that are usually defined as 'mid-to-low'. In this concentration range, the target is partly saturated.

No. Melanotan-2 is manufactured synthetically. The naturally occurring peptide in the same family is alpha-melanocyte-stimulating hormone, which the body produces as part of normal endocrine and neural signalling.
Both are synthetic analogues derived from the same parent hormone. Reference descriptions usually characterise afamelanotide as more selective for the MC1R subtype, whereas melanotan-2 is reported to interact with a wider range of melanocortin receptors.
Early studies explored melanocortin signalling and pigmentation, which later led to informal use of the label tanning peptide. That label is not a regulatory category and does not imply approved status for any purpose.
No regulatory agency has authorised melanotan-2 as a medicine for any indication. It circulates mainly as a research chemical or through unregulated channels. As a result, identity, purity and content are not independently guaranteed.
