== History == Before the development of the ELISA, the only option for conducting an immunoassay was radioimmunoassay, a technique using radioactively labeled antigens or antibodies. In radioimmunoassay, the radioactivity provides the signal, which indicates whether a specific antigen or antibody is present in the sample. Radioimmunoassay was first described in a scientific paper by Rosalyn Sussman Yalow and Solomon Berson published in 1960. As radioactivity poses a potential health threat, a safer alternative was sought. A suitable alternative to radioimmunoassay would substitute a nonradioactive signal in place of the radioactive signal. When enzymes (such as horseradish peroxidase) react with appropriate substrates (such as ABTS or TMB), a change in color occurs, which is used as a signal. However, the signal has to be associated with the presence of an antibody or antigen, which is why the enzyme has to be linked to an appropriate antibody. This linking process was independently developed by Stratis Avrameas and G. B. Pierce. Since it is necessary to remove any unbound antibody or antigen by washing, the antibody or antigen has to be fixed to the surface of the container; i.e., the immunosorbent must be prepared. A technique to accomplish this was published by Wide and Jerker Porath in 1966. == Company history == BIA Separations was formed in 1998 by a group of scientists in Ljubljana, Slovenia to commercialize a new chromatographic resin based on monolith technology. Monolithic resin was developed independently by three different laboratories in late 1980s led by Hjertén, Švec and Tennikova. The company was one of the few Slovenian companies to receive venture capital funding at that time-Horizonte Venture Management realizing the potential of monoliths made the initial investment in the company. During that year, BIA Separations released its first commercial product marketed under the trade name of CIM (Convective Interaction Media). The new technique was named HPMC (High Performance Monolith Chromatography). During the 1999 and 2000s, the company promoted this new platform technology to the scientific and business community and registered several patents. In 2007 the company headquarters moved to Villach, Austria. In 2011 BIA Separations entered a strategic partnership with Japan Synthetic Rubber Corporation, a Japanese company producing synthetic materials. === Balon === Balon (Spanish "ball", after homologue Pelota) is a hibernation factor protein found in the cold-adapted bacterium Psychrobacter urativorans. The protein was discovered accidentally by a researcher who unintentionally left a sample of P. urativorans in an ice bucket for too long, cold-shocking it, through subsequent cryo-EM scans of the organism's ribosomes. Unlike other factors, Balon can bind to the ribosome while protein production is in process. This is important for rapid response to stress because in some cells, protein production can take up to 20 minutes to complete. Balon does this by rather than physically blocking the A site of the ribosome, as other hibernation factors do, binding near to but not across the channel, allowing it to attach to the ribosome independent of whether protein production is taking place. Genetic relatives of Balon have been found in 20% of bacterial genomes catalogued in public databases, but are absent from Escherichia coli and Staphylococcus aureus, the most widely used models for cellular dormancy.

== Career == Ciulli remained in Cambridge University to conduct post-doctoral research on fragment-based drug discovery with Professor Abell and Professor Tom L. Blundell, under a College Junior Research Fellowship. Between February and June 2009, Ciulli went to Yale University as Human Frontier Science Programme visiting fellow to visit the laboratory of Professor Craig Crews before returning to Cambridge University to start his independent research career. While at Cambridge, Ciulli was the group leader in the Department of Chemistry, Director of Studies in Chemistry and BBSRC David Phillis Fellow at Christ's College. In April 2013, he took up a Readership in Chemical & Structural Biology as a principal investigator within the Division of Biological Chemistry and Drug Discovery in the University of Dundee. Ciulli was promoted as the Professor of Chemical & Structural Biology in the same division in October 2016. In 2017, he co-founded Amphista Therapeutics, a company that focuses on developing drugs based on targeted protein degradation. He was elected to the Fellowship of the Royal Society of Edinburgh in 2023.
In July 2020, the International Crisis Group (ICG) published an analysis saying that the reason for the decrease in homicides during Bukele's first year in office hypothesized that "quiet, informal understandings" between the government and the gangs. The Salvadoran government denied the ICG's allegations, and the ICG stated that it had no evidence to support the claim. In September 2020, the Salvadoran digital newspaper El Faro accused Bukele's government of conducting secret negotiations with MS-13. According to the El Faro report, the government agreed to grant MS-13 more freedom in prison in exchange for a reduction in homicides it would commit and support for Nuevas Ideas during the 2021 legislative elections. Bukele denied El Faro's allegations, posting photos on Twitter of gang members rounded up in cramped conditions from an April 2020 prison crackdown. In October 2005, work was begun by a group of Ukrainian developers to port Decay over to Windows, as a modification for both the old World Opponent Network and current Steam versions of Half-Life. The port relied on the discovery of a method that allowed Decay's PS2 game files to be converted to Windows version of the game. The port went into the beta development stage in December 2007 and was released publicly on September 23, 2008. The port was received well by the journalists in the industry; GameSpy site Planet Half-Life noted that it was a shame that an official PC version of Decay never emerged, while British journalist Alec Meer stated that it was "fantastic to have this short but sweet lost Half-Life episode on PC at last, and it even has something the PS2 version didn't—online play".

At the initiation of treatment, GnRH agonists are associated with a "flare" effect on hormone levels due to acute overstimulation of the GnRH receptor. In contrast to GnRH agonists, GnRH antagonists, such as degarelix (Firmagon) and elagolix (Orilissa), work by binding to the GnRH receptor without activating it, thereby displacing GnRH from the receptor and preventing its activation. Unlike with GnRH agonists, there is no initial surge effect with GnRH antagonists; the therapeutic effects are immediate, with sex hormone levels being reduced to castrate levels within a few days. GnRH modulators are highly effective for testosterone suppression in transgender women and have few or no side effects when sex hormone deficiency is avoided with concomitant estrogen therapy. However, GnRH modulators tend to be very expensive (typically US$10,000 to US$15,000 per year in the United States), and are often denied by medical insurance.

=== For Alzheimer's disease === Several companies are in the early stages of development and testing of this potential class of treatment. In March 2008 phase I results were reported for CoMentis Inc's candidate CTS-21166. In April 2012 Merck & Co., Inc reported phase I results for its candidate verubecestat (MK-8931). Merck began a Phase II/III trial of MK-8931 in December, 2012 estimated to be completed in July 2019. In February 2017, Merck halted its late-stage trial of verubecestat for mild to moderate Alzheimer's disease after it was reported as having "virtually no chance" of working according to an independent panel of experts. This came just three months after Eli Lilly & Co. announced its own setback with solanezumab. In September 2014 AstraZeneca and Eli Lilly and Company announced an agreement to codevelop lanabecestat (AZD3293). A pivotal Phase II/III clinical trial of lanabecestat started in late 2014, but was halted in 2018 before its planned conclusion due to poor results. Another BACE1 inhibitor that has reached phase II trials is the Eli Lilly's inhibitor LY2886721. The data on phase I trial were first presented at the Alzheimer's Association International conference in 2012. Daily dosing during 2 weeks, reduced BACE1 activity by 50–75% and CSF Aβ42 by 72% (Willis et al., 2012; Bowman Rogers and Strobel, 2013). Recently, Lilly reported that the phase II trial of LY2886721 was terminated due to liver abnormalities that were found in 4 out of 45 patients (Rogers, 2013).
